Glecaprevir, Pibrentasvir falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for Mavyret (glecaprevir and pibrentasvir). Your body partly clears glecaprevir using an enzyme called CYP3A4. The beta-carboline compounds in ayahuasca (like harmine) have been shown in lab dish studies only to slow that enzyme down. If that happened in a real person, more of the drug could linger in your blood, which might mean a bit more of the usual side effects, such as headache, tiredness, nausea, or itching.

I want to be honest about the strength of this: it's theoretical and has never actually been reported in people, so it's a maybe, not a sure thing. That said, ayahuasca carries its own serious risks and isn't something to use on your own. Please loop in your doctor or pharmacist before mixing anything new with your hepatitis C treatment.

What Is Glecaprevir and Pibrentasvir Used For?

Brand name: Mavyret

Mavyret treats acute or chronic hepatitis C virus (HCV) infection caused by genotypes 1, 2, 3, 4, 5, or 6 in adults and children 3 years and older who have no cirrhosis or have compensated cirrhosis (mild liver scarring that is still functioning).

Mavyret is also approved for adults and children 3 years and older with genotype 1 HCV who were previously treated with either an NS5A inhibitor or an NS3/4A protease inhibitor (two earlier classes of hepatitis C drugs) — but not both at the same time.
