Caffeine (OTC drug) falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for caffeine pills like Vivarin. The beta-carboline compounds in ayahuasca can slow down a liver enzyme called CYP3A4, which helps clear certain drugs. If that enzyme works slower, a drug it handles can build up and hit you harder. In theory, that could mean stronger and longer-lasting caffeine effects: a racing or pounding heart, jitteriness, anxiety, an upset stomach, and trouble sleeping.

The important part: this is based only on lab studies, not on real people, so it may not happen in your body at all. Honestly, the bigger concern is that ayahuasca itself is a powerful, risky brew that isn't recommended for self-use. Please talk with your doctor or pharmacist before combining anything with it.
