Diazepam falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for Valium (diazepam): your liver leans heavily on an enzyme called CYP3A4 to break this medicine down. The beta-carboline compounds in ayahuasca (harmine, harmaline) appear to slow that enzyme down, at least in lab studies. If that happens in your body too, diazepam could clear more slowly and build up, which may leave you feeling more sedated, drowsy, foggy, or unsteady on your feet than usual.

I want to be honest about the strength of this, though. So far this is only seen in test-tube research, not in real people, so it's more of a theoretical concern. That said, ayahuasca carries its own serious risks and isn't something to mix casually. Please check with your doctor or pharmacist before combining them.

What Is Diazepam Used For?

  • Diazepam is used to relieve anxiety and to control agitation caused by alcohol withdrawal. It is also used along with other medications to control muscle spasms and spasticity caused by certain neurological disorders such as cerebral palsy (condition that causes difficulty with movement and balance), paraplegia (inability to move parts of the body), athetosis (abnormal muscle contractions), and stiff-man syndrome (a rare disorder with muscle rigidity and stiffness). Diazepam is also used along with other medications to control seizures. Diazepam is in a class of medications called benzodiazepines. It works by calming abnormal overactivity in the brain.
