Lapatinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for Tykerb (lapatinib): your body relies on a liver enzyme called CYP3A4 to clear this drug out of your system. Ayahuasca contains plant compounds (beta-carboline alkaloids) that may slow that same enzyme down. If the enzyme works slower, lapatinib could build up higher than expected in your blood.

With more lapatinib hanging around, you might see stronger side effects, things like diarrhea, nausea, rash, fatigue, or, more seriously, effects on your heart rhythm or liver. I'll be honest though: this concern comes only from lab (test-tube) studies, and it hasn't actually been shown in people. Still, lapatinib is a serious cancer medicine, so please don't mix in ayahuasca without first talking to your oncologist or pharmacist.

About Lapatinib

What Is Lapatinib Used For?

Brand name: Tykerb

Lapatinib tablets — sold under the brand name Tykerb — are used to treat advanced or metastatic breast cancer in patients whose tumors overexpress the HER2 protein. They are always used in combination with another medicine, not alone.

Tykerb is approved in combination with capecitabine for HER2-positive advanced or metastatic breast cancer in patients who have already received prior treatment including an anthracycline, a taxane, and trastuzumab, and whose disease has progressed on trastuzumab. Tykerb is also approved in combination with letrozole for postmenopausal women with hormone receptor-positive, HER2-positive metastatic breast cancer when hormonal therapy is appropriate.
