Amphetamine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's the thing to know about combining ayahuasca with your amphetamine. The vine in ayahuasca contains compounds (harmine and harmaline) that, in lab studies, appear to slow down an enzyme called CYP2D6, which is one of the routes your body uses to clear amphetamine. If that enzyme gets blocked, more amphetamine can linger in your system, which could mean a stronger stimulant effect: think a racing heart, higher blood pressure, jitteriness, trouble sleeping, or feeling anxious and wired.

Now, I want to be honest about the evidence. This is based on test-tube and animal research, not real people, so it's theoretical. But there's a bigger separate concern: ayahuasca also acts as an MAOI, and mixing MAOIs with stimulants like amphetamine can be genuinely dangerous. Please talk with your doctor or pharmacist before going anywhere near this combination.

Amphetamine treats Attention Deficit Hyperactivity Disorder (ADHD) in patients 6 years and older. Adzenys XR-ODT, Dyanavel XR, and the amphetamine extended-release orally disintegrating tablet formulations are specifically approved for ADHD in this age group.

Amphetamine Sulfate tablets and Evekeo (amphetamine sulfate tablets) are also approved for narcolepsy (a sleep disorder causing uncontrollable daytime sleepiness) and as a short-term aid in a calorie-restricted weight-loss program for exogenous obesity (weight gain from external factors like overeating) in patients who have not responded to other approaches.
