Glecaprevir, Pibrentasvir falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Adrafinil interacts with.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to weakly induce CYP3A4 activity.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the concern with pairing adrafinil and Mavyret (glecaprevir and pibrentasvir). Once your liver turns adrafinil into modafinil, that modafinil can mildly speed up an enzyme called CYP3A4. Since Mavyret is partly handled by that same enzyme, revving it up could lower the amount of medication in your blood. For a hepatitis C treatment, that matters, because you need steady, full drug levels to fully clear the virus. Weaker levels could mean the treatment doesn't work as well as it should.
To be fair, this is a theoretical concern based on how these drugs behave, not strong real-world proof, and modafinil is only a weak inducer. Still, given the stakes of curing hepatitis C, please run this by your doctor or pharmacist before combining them.

What Is Glecaprevir and Pibrentasvir Used For?
Brand name: Mavyret
Mavyret treats acute or chronic hepatitis C virus (HCV) infection caused by genotypes 1, 2, 3, 4, 5, or 6 in adults and children 3 years and older who have no cirrhosis or have compensated cirrhosis (mild liver scarring that is still functioning).
Mavyret is also approved for adults and children 3 years and older with genotype 1 HCV who were previously treated with either an NS5A inhibitor or an NS3/4A protease inhibitor (two earlier classes of hepatitis C drugs) — but not both at the same time.
