Theoretically, adrafinil might reduce the levels and clinical effects of CYP1A2 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to weakly induce CYP1A2 activity in vitro.

Here's the short version: your body turns adrafinil into modafinil, and modafinil may gently speed up a liver enzyme called CYP1A2. Naproxen is partly cleared by that same enzyme. So in theory, taking adrafinil could nudge your body to break down naproxen a little faster, which might make it a touch less effective at easing your pain, swelling, or joint stiffness.

I want to be honest with you, though: this concern is theoretical. It comes from lab studies, not from real patients, and the effect on naproxen looks weak. Most people likely wouldn't notice a difference. Naproxen isn't a prodrug, so this won't make it more toxic. Adrafinil's bigger worry is its strain on the liver. Please check with your pharmacist or doctor before combining them.

Here's the gist for naproxen. Your liver uses an enzyme called CYP2C9 to help clear naproxen out of your system. Adrafinil turns into modafinil in your body, and lab studies suggest modafinil may slow that same enzyme down a bit. If it does, naproxen could linger a little longer and its effects (and side effects) could be slightly stronger.

For naproxen, that might mean more stomach upset, heartburn, or in some cases stomach irritation or bleeding, plus fluid retention. Honestly, this is a theoretical concern based on test-tube data, not real-world reports, so I wouldn't lose sleep over it. Still, since adrafinil itself can stress the liver, it's worth running both past your pharmacist or doctor first.

About Naproxen

What Is Naproxen Used For?

Brand names: Naprosyn, Naprelan, Anaprox, Anaprox DS, Naproxyn, Navalbine, Naproxen, Naprosyn EC, Napro-Na, Aleve, Arthroxen, Synflex, EC-Naprosyn

Naproxen tablets, naproxen sodium tablets, and naproxen oral suspension are used to relieve the signs and symptoms of rheumatoid arthritis, osteoarthritis, ankylosing spondylitis, and polyarticular juvenile idiopathic arthritis. They are also used for tendonitis, bursitis, and acute gout, and for managing pain and primary dysmenorrhea (menstrual cramps).

Naproxen Sodium Controlled-Release Tablets are indicated for the same conditions plus the relief of mild to moderate pain. Several over-the-counter naproxen sodium products are indicated for the temporary relief of minor aches and pains from headache, backache, muscular aches, toothache, menstrual cramps, minor arthritis pain, and the common cold, and to temporarily reduce fever.
