Why this applies to Memantine, Donepezil

Memantine, Donepezil falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Namzaric combines memantine and donepezil, and donepezil is partly cleared by a liver enzyme called CYP2D6. The beta-carboline compounds in ayahuasca (harmine and harmaline) may block that enzyme. If they do, donepezil could build up a bit, which might mean stronger side effects like nausea, diarrhea, slow heart rate, dizziness, muscle cramps, or trouble sleeping.

I want to be honest about the evidence here: this is only seen in lab (test tube) studies, not in real people, so it stays mostly theoretical. That said, ayahuasca is a potent, risky brew on its own and is not something to mix casually. Please loop in your doctor or pharmacist before combining anything with Namzaric.

Here's the basic idea with Namzaric (which combines memantine and donepezil) and ayahuasca. The beta-carboline compounds in ayahuasca can slow down a liver enzyme called CYP3A4, and donepezil leans on that enzyme to get cleared from your body. If that pathway gets blocked, donepezil could build up a bit and you might feel its side effects more strongly, things like nausea, diarrhea, dizziness, slow heartbeat, or trouble sleeping.

That said, I want to keep this in perspective. This was only seen in lab dishes, not in real people, so it's a theoretical concern. Honestly, the much bigger issue is that ayahuasca itself can be genuinely dangerous. Please loop in your doctor or pharmacist before mixing anything like this.
