Why this applies to Chlorpheniramine

Chlorpheniramine falls under SEROTONERGIC DRUGS — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase levels of serotonin, which may have additive effects with serotonergic drugs.
In vitro and animal research shows that harmaline and harmine, constituents of ayahuasca, strongly and reversibly inhibit monoamine oxidase A and weakly inhibit monoamine oxidase B. The dimethyltryptamine and tetrahydroharmine constituents of ayahuasca have also demonstrated serotonergic activity. A case of serotonin syndrome characterized by tremors, sweating, shivering, confusion, nausea, vomiting, and disorientation has been reported in a 36-year-old male who consumed 100 mL of ayahuasca while taking fluoxetine 20 mg daily. Combining serotonergic drugs with ayahuasca might increase the risk of serotonergic side effects including serotonin syndrome and cerebral vasoconstrictive disorders.

Here's the thing to keep in mind: ayahuasca contains compounds that act like strong MAO inhibitors and can push up serotonin levels in your body. Chlorpheniramine is mainly an antihistamine, but it also has mild serotonin-related activity, so on paper combining the two could add together and raise your risk of too much serotonin.

I'll be straight with you, this concern is theoretical and based on how these substances behave, not on real cases with chlorpheniramine specifically. Still, serotonin syndrome (think shivering, sweating, tremor, confusion, racing heart, nausea) is serious, and ayahuasca itself is risky and illegal in many places. Please don't mix them on your own. Talk with your pharmacist or doctor first so they can look at your full list of medicines.

Chlorpheniramine also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for chlorpheniramine. Your liver uses an enzyme called CYP3A4 to help clear this antihistamine. The beta-carboline compounds in ayahuasca may slow that enzyme down, at least in lab studies. If that slowdown happened in real life, chlorpheniramine could linger longer and build up, making its usual side effects stronger, things like drowsiness, dry mouth, blurry vision, constipation, and feeling foggy or dizzy.

That said, this is theoretical and only seen in test-tube research, not in actual people, so I wouldn't panic. Honestly, the bigger concern with ayahuasca is its powerful MAOI activity and the serious risks of using it at all. Please loop in your doctor or pharmacist before mixing anything with it.

Chlorpheniramine is used to temporarily relieve common allergy symptoms caused by hay fever (allergic rhinitis) or other upper respiratory allergies. These include runny nose, sneezing, itchy or watery eyes, and itching of the nose or throat.

These uses are supported across multiple products — including tablets (such as Aller-chlor, Allergy, Allergy Relief, Allergy Time, Anti Allergy, and Chlorpheniramine Maleate), extended-release tablets (Allergy Relief and Chlorphen-12), the oral solution Ed Chlorped Jr, and the inhalers ClorRelief and ClorRelief KIDs.
