Here's the gist with Copavin (codeine plus papaverine) and rauwolscine. This isn't about how your body breaks the drug down. Instead, it's about seizure risk. Some medications, including ones in this group, can slightly lower the threshold for a seizure. The concern here is that rauwolscine, a strong stimulant alkaloid, may nudge that threshold lower too, so stacking them together could in theory add up.

I'll be straight with you: this is a theoretical caution based only on animal research, not human cases, so the real-world risk is likely small for most people. Still, since rauwolscine also revs up heart rate, blood pressure, and anxiety, it's worth running this combo by your pharmacist or doctor before mixing them.

Theoretically, rauwolscine might increase levels of drugs metabolized by CYP2D6.
Rauwolscine is structurally related to yohimbine. In vitro research shows that yohimbine inhibits CYP2D6 enzyme activity.

Here's the wrinkle with this combo. Codeine is what we call a prodrug, meaning it doesn't do much on its own. Your body uses an enzyme called CYP2D6 to turn codeine into morphine, which is the part that actually relieves pain and makes you drowsy. Rauwolscine is a cousin of yohimbine, and in lab studies yohimbine appears to slow down that same CYP2D6 enzyme.

So in theory, rauwolscine could reduce how well codeine works, leaving you with weaker pain relief. I'll be honest though, this is based only on test-tube data, not real people, so it's far from certain. There's also a separate concern worth mentioning: rauwolscine is a stimulant that raises heart rate and blood pressure, which doesn't pair nicely with a sedating opioid. Please run this past your pharmacist or doctor before combining them.
