Here is the interaction we found between these two, with the evidence behind it.
CNS DEPRESSANTS
Why this applies to Ergotamine, Belladonna, Phenobarbital
Ergotamine, Belladonna, Phenobarbital falls under CNS DEPRESSANTS — the group Saffron interacts with.
Clinical research shows that taking saffron extract 60 mg orally daily for 26 weeks can cause drowsiness and sedation. Animal research suggests that adding saffron to hexobarbital further increases sleeping and slows motor activity.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
This combination product contains phenobarbital, a barbiturate that already slows down your brain and nervous system. Saffron, when taken in concentrated supplement doses, may add a little extra to that calming, sleepy effect. So the worry here is simple: stacking the two could leave you more drowsy, foggy, dizzy, or off-balance than usual.
I want to be honest that this is a theoretical concern based on how these things work, plus some animal studies, not strong human proof. Food amounts of saffron in cooking are very unlikely to cause trouble. The caution is really about daily saffron capsules. If you take this medication, just check with your pharmacist or doctor before adding a saffron supplement, and don't drive until you know how you feel.

From our pharmacist’s overview on the Ergotamine, Belladonna, Phenobarbital page
“Sedation stacking is the risk I would put first with ergotamine, belladonna, and phenobarbital (Bellamine, Bellaspas). Phenobarbital is a barbiturate, sedating on its own, and 78 of the ingredients on this page deepen that drowsiness, alcohol most seriously of all.”

About Saffron
What is Saffron?
- Depression and low mood
- Anxiety
- PMS symptoms
- Memory and cognition
- Eye health (age-related macular degeneration)
Saffron is a costly spice that has shown promise in early studies for mild-to-moderate depression and some mood and PMS symptoms, but most research uses small trials, so results are not definitive. It is generally well tolerated in food and small supplement amounts, but high doses can be harmful and it should be avoided in pregnancy. Talk to your pharmacist or doctor before using saffron supplements.
