Why this applies to Adagrasib

Adagrasib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Sulforaphane interacts with.

Theoretically, sulforaphane might increase the levels and effects of CYP3A4 substrates.
Animal and in vitro research shows that sulforaphane inhibits CYP3A4 enzyme activity and downregulates its expression. This interaction has not been reported in humans.

Adagrasib (Krazati) is a cancer pill that your liver clears using an enzyme called CYP3A4. In lab dishes and animal studies, sulforaphane has been shown to slow that same enzyme down. If that were to happen in people, your body might break down adagrasib more slowly, letting it build up. With higher levels you could see more of its usual side effects, things like nausea, diarrhea, fatigue, or changes in heart rhythm (QT prolongation).

The important catch: this is only theoretical right now and has never actually been reported in humans. Eating broccoli and other veggies is very unlikely to cause a problem. A concentrated sulforaphane supplement is a different story, so please run it by your oncology pharmacist or doctor before starting one.

About Adagrasib

What Is Adagrasib Used For?

Brand name: Krazati

Krazati (adagrasib) is used to treat adults with locally advanced or metastatic non-small cell lung cancer (NSCLC) that carries a specific genetic change called a KRAS G12C mutation. It is used as a single agent for NSCLC after at least one prior treatment, and patients must be confirmed to have this mutation by an FDA-approved test.

Krazati is also used in combination with another drug called cetuximab to treat adults with KRAS G12C-mutated locally advanced or metastatic colorectal cancer (CRC) who have already been treated with specific prior chemotherapy regimens. Both uses were approved under the FDA's accelerated approval program based on tumor response data, and continued approval may depend on results from confirmatory trials.
