Why this applies to Everolimus

Everolimus falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Sulforaphane interacts with.

Theoretically, sulforaphane might increase the levels and effects of CYP3A4 substrates.
Animal and in vitro research shows that sulforaphane inhibits CYP3A4 enzyme activity and downregulates its expression. This interaction has not been reported in humans.

Here's the gist for everolimus (Afinitor, Zortress): your body relies on an enzyme called CYP3A4 to break it down. In lab and animal studies, sulforaphane appears to slow that enzyme down. If that happened in people, everolimus could build up to higher levels than expected, which may mean more side effects like mouth sores, fatigue, low blood counts, high blood sugar, or higher risk of infection.

I want to be honest about the strength of this, though. This effect has only been seen in test tubes and animals, never confirmed in humans, so it's theoretical for now. Eating broccoli at dinner is very unlikely to matter. A concentrated sulforaphane supplement is the real question, so please run it by your oncologist or pharmacist before starting one.

About Everolimus

What Is Everolimus Used For?

Brand names: Certican, Afinitor, Zortress, Afinitor Disperz

Everolimus is used to treat several serious conditions. In cancer, it is prescribed for advanced hormone receptor-positive, HER2-negative breast cancer (in postmenopausal women), certain neuroendocrine tumors of the pancreas, GI tract, or lung, and advanced renal cell carcinoma (kidney cancer) when other treatments have stopped working.

It is also used for people with tuberous sclerosis complex (TSC) — a genetic condition that causes benign tumors to grow throughout the body. In TSC, it treats kidney tumors called angiomyolipomas, brain tumors called SEGA, and seizures. In kidney transplant patients, it helps prevent the body from rejecting the new organ.
