Why this applies to Glasdegib

Glasdegib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Sweet Annie interacts with.

Sweet Annie may alter plasma levels and clinical effects of drugs metabolized by CYP3A4.
In vitro research shows that the Sweet Annie constituent artemisinin induces CYP3A4, possibly increasing CYP3A4 activity by 1.9-fold. However, Sweet Annie extract seems to inhibit the activity of CYP3A4 in vitro, suggesting that other constituents of Sweet Annie play a role in its effects on the overall activity of this enzyme. More information is needed to determine whether taking Sweet Annie extract affects the metabolism of CYP3A4 substrates.

Here's the situation with Sweet Annie and Glasdegib (Daurismo). Your body clears Glasdegib using a liver enzyme called CYP3A4, and Sweet Annie may bump into that same enzyme. The tricky part is the lab data points in two directions: one compound (artemisinin) may speed the enzyme up, while the whole-plant extract may slow it down. So we honestly can't say for sure which way Glasdegib levels would move.

If Glasdegib built up, you could see more of its known effects, like nausea, fatigue, muscle cramps, or changes in your heart's QT rhythm. If levels dropped, the leukemia treatment might work less well. Because this is a serious cancer drug, please don't combine them without checking with your oncology team or pharmacist first. The evidence here is only lab-based, so this is more of a caution than a proven problem.

What Is Glasdegib Used For?

Brand name: Daurismo

Daurismo (glasdegib) is approved to treat newly-diagnosed acute myeloid leukemia (AML) — a fast-growing cancer of the blood and bone marrow. It is used specifically in adults who are 75 years of age or older, or in adults of any age whose other health conditions make intensive induction chemotherapy too risky.

Daurismo is not used alone. It is always given together with low-dose cytarabine, a separate chemotherapy medicine, as part of a 28-day treatment cycle.
