Here's the concern with pairing Sweet Annie and Sotorasib (Lumakras). This isn't about one changing how the other is broken down. Instead, both can be tough on your liver. Sotorasib is already known to raise liver enzymes and, in some people, cause liver inflammation. Sweet Annie has been linked to liver irritation too, so taking them together could theoretically add up to more strain on that organ.

To be honest, this is a theoretical, mostly anecdotal caution, not something proven in studies. Still, since your care team likely checks your liver labs while you're on Sotorasib, please loop in your oncologist or pharmacist before adding Sweet Annie. Watch for yellowing skin or eyes, dark urine, belly pain, or unusual tiredness, and report those right away.

Sotorasib also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

Sweet Annie may alter plasma levels and clinical effects of drugs metabolized by CYP3A4.
In vitro research shows that the Sweet Annie constituent artemisinin induces CYP3A4, possibly increasing CYP3A4 activity by 1.9-fold. However, Sweet Annie extract seems to inhibit the activity of CYP3A4 in vitro, suggesting that other constituents of Sweet Annie play a role in its effects on the overall activity of this enzyme. More information is needed to determine whether taking Sweet Annie extract affects the metabolism of CYP3A4 substrates.

Here's the situation with Sweet Annie and Lumakras (sotorasib). Your sotorasib is cleared from your body by a liver enzyme called CYP3A4. Sweet Annie can tug on that enzyme, but honestly the lab data points both ways: one compound in it (artemisinin) may speed the enzyme up, while the whole-plant extract seems to slow it down. So we can't say for sure which direction would win in a real person.

Why this matters: if the enzyme speeds up, your sotorasib could clear faster and work less well against your cancer. If it slows down, levels could climb and you might see more side effects like diarrhea, nausea, fatigue, or liver irritation. This is only test-tube evidence, not proven in people, but because sotorasib is a cancer drug I'd play it safe. Please check with your oncologist or pharmacist before adding Sweet Annie.

Sweet Annie (Artemisia annua) is the source of artemisinin, a compound used in prescription antimalarial drugs. While the purified drug is well studied for malaria, the herb itself as a supplement is not proven for the wide range of conditions people use it for, and self-treating malaria with the raw herb is dangerous. About Sotorasib

What Is Sotorasib Used For?

Brand name: Lumakras

Lumakras is used to treat adults with KRAS G12C-mutated locally advanced or metastatic non-small cell lung cancer (NSCLC) who have already received at least one prior cancer treatment. This use was approved under the FDA's accelerated approval pathway based on tumor response rates, and continued approval may depend on results from confirmatory trials.

Lumakras is also used in combination with panitumumab to treat adults with KRAS G12C-mutated metastatic colorectal cancer (mCRC) who have previously received certain chemotherapy regimens. In both cases, patients must first be confirmed to carry the KRAS G12C mutation using an FDA-approved test.
