Leniolisib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Tree Turmeric interacts with.

Theoretically, tree turmeric might increase the levels and clinical effects of drugs that are substrates of CYP3A4.
In vitro research and preliminary clinical evidence suggest that berberine, a constituent of tree turmeric, moderately inhibits CYP3A4.

Here's the short version: your body clears leniolisib (Joenja) using a liver enzyme called CYP3A4. Berberine, the active piece of tree turmeric, appears to slow that same enzyme down. When the cleanup crew works slower, leniolisib can build up higher than intended, which may mean stronger effects and a greater chance of side effects like headache, diarrhea, nausea, or feeling more tired than usual.

This is rated as a probable, moderate interaction, though the leniolisib-specific piece is more theoretical since it's based on how berberine behaves with CYP3A4 in general. Small culinary amounts are unlikely to be the concern, but concentrated berberine supplements could be. Please don't stop or change anything on your own. Just run the combination past your pharmacist or doctor first.

What Is Leniolisib Used For?

Brand name: Joenja

Joenja (leniolisib) is approved to treat activated phosphoinositide 3-kinase delta syndrome, commonly called APDS — a rare genetic immune disorder in which certain immune cells (B and T cells) don't work properly. It is approved for adults and pediatric patients who are at least 12 years old.

APDS is caused by gene variants that make a specific enzyme (PI3K-delta) overactive, leading to immune system dysfunction. Joenja is the brand name for leniolisib in tablet form, taken by mouth.
