Tree Turmeric and Amitriptyline

These two interact in 4 distinct ways. Each one is explained separately below.

Why Are There 4 Interactions?

Tree Turmeric interacts with whole categories of medication. Amitriptyline falls into 4 of those categories, so it can interact with Tree Turmeric in 4 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).

Moderate

CNS DEPRESSANTS

1 of 4

Why this applies to Amitriptyline

Amitriptyline falls under CNS DEPRESSANTS — the group Tree Turmeric interacts with.

Theoretically, use of tree turmeric along with CNS depressants might increase the risk of additive therapeutic and adverse effects.
Animal research suggests that berberine, a constituent of tree turmeric, can have sedative effects.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the simple version: amitriptyline already tends to make people drowsy, especially when you first start it or at bedtime. Tree turmeric (the berberine in it) has shown sedative effects in animal studies, so the worry is that pairing the two could leave you feeling extra sleepy, groggy, or foggy. Think more pronounced versions of amitriptyline's usual side effects, like daytime tiredness or trouble concentrating.

I want to be honest about the evidence, though: this is theoretical and based only on lab and animal research, not real human reports, so it may never cause you any trouble. Still, if you decide to try tree turmeric, mention it to your pharmacist or doctor first, and go easy until you know how the combination affects you.

CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES

2 of 4

Amitriptyline also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.

Theoretically, tree turmeric might increase the levels and clinical effects of drugs metabolized by CYP2D6.
In vitro research and preliminary clinical evidence suggest that berberine, a constituent of tree turmeric, can inhibit CYP2D6.
Severity Moderate Likelihood Possible Evidence B

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the gist for amitriptyline (Elavil): your body relies on a liver enzyme called CYP2D6 to break it down and clear it out. The berberine in tree turmeric appears to slow that enzyme down. When the cleanup crew works slower, amitriptyline can build up a bit higher than usual.

Practically, that could mean stronger side effects like drowsiness, dry mouth, constipation, blurry vision, dizziness when standing, or a faster heartbeat. To be fair, this is mostly based on lab work and some early, lower quality human data, so it's a possible concern, not a sure thing. Concentrated berberine supplements are more likely to matter than tiny culinary amounts. Please don't change anything on your own, just check with your pharmacist or doctor first.

CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES

3 of 4

Amitriptyline also falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES.

Theoretically, tree turmeric might increase the levels and clinical effects of drugs metabolized by CYP2C9.
Preliminary clinical evidence suggests that berberine, a constituent of tree turmeric, can inhibit CYP2C9.
Severity Moderate Likelihood Possible Evidence B

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the gist: the berberine in tree turmeric can slow down a liver enzyme called CYP2C9, which is one of the pathways your body uses to break down amitriptyline. When that enzyme is partly blocked, amitriptyline can clear out of your system a little slower and build up. That means you might feel more of its effects, including the side effects this medicine is known for, like drowsiness, dry mouth, constipation, blurred vision, dizziness when standing up, or a faster heartbeat.

I want to be honest about the evidence here, though. This is mostly theoretical and based on early, lower-quality research, so it may never cause a noticeable problem for you. Still, since amitriptyline can be touchy at higher levels, please check with your pharmacist or doctor before pairing the two rather than guessing.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

4 of 4

Amitriptyline also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

Theoretically, tree turmeric might increase the levels and clinical effects of drugs that are substrates of CYP3A4.
In vitro research and preliminary clinical evidence suggest that berberine, a constituent of tree turmeric, moderately inhibits CYP3A4.
Severity Moderate Likelihood Probable Evidence A

Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the gist for these two together. Your body uses a liver enzyme called CYP3A4 to help clear amitriptyline. Berberine, the active piece of tree turmeric, can moderately slow that enzyme down. When the cleanup slows, more amitriptyline can linger in your system, which may nudge its effects and side effects upward.

For amitriptyline that can mean more drowsiness, dry mouth, constipation, dizziness, blurred vision, or a faster heartbeat. The interaction is partly theoretical, but the evidence that berberine blocks this enzyme is fairly solid, so it's worth respecting. Concentrated berberine supplements matter more than a sprinkle in cooking. Please don't change anything on your own. Just loop in your pharmacist or doctor so they can watch for these effects.
