Allyzyme contains 20 active ingredients, most of them digestive enzymes. The enzyme blend includes amylase (for starch), papain (for protein), bromelain (for protein), lipase (for fat), lactase (for lactose), cellulase, peptidase, invertase, glucoamylase, alpha-galactosidase, xylanase, pectinase, protease variants, acid maltase, and CereCelase.
The formula also includes herbal extracts: artichoke leaf extract and gentian root extract. These are inactive ingredients: natural vegetable capsules, silicon dioxide, and magnesium stearate.
Does it work?
Strong evidence
Lactase is the only ingredient here rated effective for its purpose—it works for lactose intolerance. Artichoke leaf extract is possibly effective for high cholesterol (hyperlipidemia) and indigestion (dyspepsia).
For the other active ingredients—papain, bromelain, lipase, protease enzymes, gentian root, and the remaining enzyme components—the evidence we hold isn't established. That doesn't mean they don't work; it means the research isn't strong enough to rate them one way or the other.
How safe is it?
Well-documented data
Most of these ingredients are generally well tolerated at typical doses. The biggest caution is allergic reactions: papain, bromelain, proteases, artichoke, and gentian can all trigger allergies in sensitive people, ranging from mild (itching, rash, stomach upset) to serious (anaphylaxis with artichoke in inulin-sensitive individuals, or respiratory symptoms with inhaled enzyme dust).
Papain at high doses may rarely cause severe gastritis or even esophageal damage. Bromelain may cause diarrhea, gas, or stomach upset.
Artichoke and gentian can cause abdominal pain, diarrhea, nausea, and flatulence. If you're pregnant, papain and bromelain are best avoided; artichoke and gentian are not recommended; and lipase and lactase lack enough data—talk to your doctor.
If you're breastfeeding, the safety data is thin across most ingredients; check with your pharmacist or doctor first.

The most serious concern is papain, which may increase the effects and side effects of warfarin (a blood thinner). In one case, a patient stable on warfarin developed an elevated clotting ratio after taking a supplement containing papain from papaya.

Bromelain may increase bleeding risk if you're taking anticoagulants or antiplatelet drugs like aspirin or clopidogrel (Plavix). It may also slightly increase tetracycline antibiotic levels, though human evidence is limited.
Artichoke leaf extract may lower blood pressure and blood sugar, so it could amplify the effects of antihypertensive drugs and diabetes medications if you're on them. It may also raise levels of drugs your liver breaks down via the CYP2C19 and CYP2B6 pathways.
Gentian root extract may increase the blood-pressure-lowering effect of antihypertensive drugs.

Artichoke leaf extract
Antidiabetes Drugs
Theoretically, artichoke leaf extract may increase the risk of hypoglycemia when taken with antidiabetes drugs.
A meta-analysis of small clinical studies shows that taking artichoke leaf extract for 8-12 weeks can modestly reduce fasting plasma glucose when compared with placebo.
Antihypertensive Drugs
Theoretically, artichoke leaf extract may increase the risk of hypotension when taken with antihypertensive drugs.
A meta-analysis of small clinical studies in patients with hypertension shows that taking artichoke can reduce systolic blood pressure by around 3 mmHg and diastolic blood pressure by around 2 mmHg when compared with placebo.
Cytochrome P450 2B6 (Cyp2B6) Substrates
Theoretically, artichoke might increase serum levels of drugs metabolized by CYP2B6.
In vitro research shows that artichoke leaf extract inhibits CYP2B6 activity. However, this interaction has not been reported in humans.
Cytochrome P450 2C19 (Cyp2C19) Substrates
Theoretically, artichoke might increase serum levels of drugs metabolized by CYP2C19.
In vitro research shows that artichoke leaf extract inhibits CYP2C19 activity. However, this interaction has not been reported in humans.
Gentian root extract
Antihypertensive Drugs
Theoretically, taking gentian with antihypertensive drugs might increase the risk of hypotension.
In vitro research shows that gentian can cause vasodilation and lower blood pressure.
Bromelain
Anticoagulant/Antiplatelet Drugs
Bromelain may have antiplatelet effects and may increase the risk of bleeding if used with anticoagulant or antiplatelet drugs.
There is one case report of a patient experiencing minor bruising while taking bromelain with naproxen. Bromelain is thought to have antiplatelet activity. Whether this interaction is of concern with topical bromelain is unclear. Interference with coagulation of burn wounds has been reported in a patient receiving bromelain-based enzymatic debridement. However, observational research has found that topical bromelain debridement is not associated with increases or decreases in laboratory markers of coagulation when compared with surgical debridement.
Tetracycline Antibiotics
Theoretically, bromelain might increase levels of tetracycline antibiotics.
Laboratory research suggests that bromelain might increase the absorption of tetracycline antibiotics. However, a study in healthy adults reported no difference in tetracycline plasma levels when a 500 mg dose was taken with or without bromelain 80 mg.
Papain
Warfarin (Coumadin)
Theoretically, papain might increase the effects and side effects of warfarin.
In one case report, a patient previously stable on warfarin was found to have an international normalization ratio (INR) of 7.4, which was attributed to ingestion of a supplement containing papain from papaya extract.
