L-glutamine is generally well tolerated in healthy adults. The most common side effects are gastrointestinal — belching, bloating, constipation, diarrhea, flatulence, nausea, and vomiting.

Headache and musculoskeletal pain have also been reported. At very high doses, dizziness and mania have occurred in rare cases, particularly in people with bipolar disorder; these resolved when the dose was lowered.
