Why this applies to Glasdegib

Glasdegib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Aegeline interacts with.

Aegeline inhibits cytochrome P450 3A4 (CYP3A4) activity in vitro. So far, this interaction has not been reported in humans. Theoretically, aegeline may increase levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others. Use aegeline cautiously or avoid in patients taking these drugs.

Your body relies on a liver enzyme called CYP3A4 to break down glasdegib (Daurismo). In lab studies, aegeline appears to slow that same enzyme down. If that happened in people, glasdegib could clear more slowly and build up in your system, which may lead to stronger effects and more side effects, like muscle cramps, nausea, hair thinning, taste changes, or changes in your heart rhythm (QT prolongation) that your cancer team already watches closely.

Here's the important context: this is only a theoretical concern from test-tube research, not something reported in real patients. Even so, aegeline itself has been tied to serious liver injury and is best avoided. Since glasdegib is a serious leukemia medicine, please don't add any supplement without first checking with your oncologist or pharmacist.

Aegeline is a compound originally identified in the bael tree (Aegle marmelos) that gained attention because it was added to some weight-loss and workout supplements. It is best known for being linked to serious liver injury in a product called OxyELITE Pro, and U.S. regulators warned against products containing it. There is no good evidence it safely helps with weight loss, and it should generally be avoided.

What Is Glasdegib Used For?

Brand name: Daurismo

Daurismo (glasdegib) is approved to treat newly-diagnosed acute myeloid leukemia (AML) — a fast-growing cancer of the blood and bone marrow. It is used specifically in adults who are 75 years of age or older, or in adults of any age whose other health conditions make intensive induction chemotherapy too risky.

Daurismo is not used alone. It is always given together with low-dose cytarabine, a separate chemotherapy medicine, as part of a 28-day treatment cycle.
