Methadone falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Clove interacts with.

Theoretically, concomitant use of clove may increase levels of drugs metabolized by CYP2D6.
In vitro research shows that eugenol, the principal constituent of clove, can inhibit CYP2D6 in a dose-dependent manner. This effect has not been reported in humans.

Here's the gist for clove and methadone. Lab studies suggest that eugenol, the main compound in clove, can slow down a liver enzyme called CYP2D6, which is one of the pathways your body uses to clear methadone. In theory, if that enzyme is blocked, methadone could build up a bit, which might mean stronger effects like extra drowsiness, slowed breathing, constipation, or feeling overly sedated.

That said, this has only been seen in test tubes, not in real people, so it's far from proven. The small amount of clove you'd get from cooking is very unlikely to matter. Concentrated clove oil or supplements are a different story. Since methadone is a powerful medicine, please run any regular clove use past your pharmacist or doctor first.

Here's the simple version: your body relies on a liver enzyme called CYP3A4 to help break down methadone. In lab dishes, eugenol (the main oil in clove) appears to slow that enzyme down. If that were to happen in real life, methadone could clear more slowly and build up, which means stronger opioid effects like extra drowsiness, slowed breathing, confusion, or constipation.

I want to be honest about the evidence, though. This is only seen in test-tube studies, not in actual people, so it's theoretical. Sprinkling clove in your cooking is very unlikely to matter. Concentrated clove oil or capsules are the bigger question mark. Since methadone is a serious medication, please check with your pharmacist or doctor before adding clove supplements.

Food amounts are generally safe, but concentrated clove oil and supplements can be irritating or toxic if overused, and human evidence for most health claims is limited. About Methadone

What Is Methadone Used For?

Brand name: Dolophine

Methadone treats severe and persistent pain that requires daily, around-the-clock, long-term opioid treatment and cannot be managed adequately with other options. It is available in several forms for this use, including oral solution, oral tablets, oral concentrate, and injectable forms (intravenous, intramuscular, and subcutaneous). Injectable methadone is not approved for outpatient pain treatment — it is reserved for hospitalized patients unable to take oral medication.

Methadone is also used for the detoxification and maintenance treatment of opioid addiction (dependence on heroin or other morphine-like drugs). Oral forms — including tablets, dispersible tablets (such as Methadose Dispersible and DISKETS), and oral concentrates (such as Methadone Hydrochloride Intensol™, Methadose, and others) — are approved for this use. By law, methadone for opioid addiction treatment must be dispensed only by certified opioid treatment programs. Injectable methadone is not approved for outpatient addiction treatment.
