Acetaminophen, Dichloralantipyrine, Isometheptene falls under CYTOCHROME P450 2E1 (CYP2E1) SUBSTRATES — the group Diosmin interacts with.
Chlorzoxazone is metabolized by CYP2E1 enzymes. A pharmacokinetic study in humans shows that taking diosmin (Venex 500) 500 mg daily for 9 days prior to oral administration of chlorzoxazone (Paraflex 250) 250 mg increases blood levels of chlorzoxazone by 34% and decreases chlorzoxazone clearance by 40%. It is speculated that diosmin reduces the metabolism of chlorzoxazone by inhibiting CYP2E1.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
This combination product (often sold as Midrin or Migquin) blends acetaminophen with a mild sedative and isometheptene for tension or migraine headaches. The piece that connects to diosmin here is the acetaminophen. Your liver uses several pathways to clear acetaminophen, and one minor pathway, an enzyme called CYP2E1, can turn a small portion of it into a byproduct that is hard on the liver.
In theory, diosmin may slow CYP2E1 a bit. Honestly, that is more likely to nudge acetaminophen away from that harmful byproduct, not toward more side effects. This is theoretical only, based on a study with a different drug, so I would not lose sleep over it. Just keep acetaminophen within recommended daily limits and check with your pharmacist if you take it regularly.

Diosmin is a citrus-derived flavonoid most often used for vein-related problems such as chronic venous insufficiency and hemorrhoids, and it is usually combined with a related flavonoid called hesperidin. Some studies suggest it may help relieve leg swelling and hemorrhoid symptoms, but quality varies.
