CHLORZOXAZONE (Parafon Forte, Paraflex)

Theoretically, diosmin might increase the levels and clinical effects of chlorzoxazone.
A pharmacokinetic study in humans shows that taking diosmin (Venex 500) 500 mg daily for 9 days prior to oral administration of chlorzoxazone 250 mg increases blood levels of chlorzoxazone by 53% and decreases chlorzoxazone clearance by 40%. It is speculated that diosmin reduces the metabolism of chlorzoxazone by inhibiting cytochrome P450 2E1 (CYP2E1).

Here is what's going on if you take diosmin along with your muscle relaxer chlorzoxazone. Chlorzoxazone is cleared from your body by a liver enzyme called CYP2E1, and diosmin appears to slow that enzyme down. In a small human study, people taking diosmin had about 53% higher chlorzoxazone levels and cleared the drug roughly 40% slower. In plain terms, more of the medicine sticks around longer.

What that can feel like is more of chlorzoxazone's usual effects: extra drowsiness, dizziness, lightheadedness, or feeling unusually weak or sluggish. None of this means you can't use both, but it's worth a quick chat with your pharmacist or doctor, especially if you notice you feel groggier than normal. Don't change either one on your own.
