Tasimelteon falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Grape interacts with.
A small pharmacokinetic study in healthy adults shows that ingestion of 200 mL of grape juice decreases phenacetin plasma levels. This is thought to be due to induction of CYP1A2.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for Hetlioz (tasimelteon). Your body relies on a liver enzyme called CYP1A2 to break this medicine down. There's a small, lower quality study suggesting grape juice may actually speed that enzyme up a bit. If that holds true, your body could clear tasimelteon a little faster, which might leave it working less well for resetting your sleep-wake cycle.
In plain terms, the worry is weaker benefit, not a dangerous spike. And honestly, this is mostly theoretical and based on weak evidence, so I wouldn't panic over an occasional glass of grape juice or eating grapes. If you're thinking about a concentrated grape seed extract supplement, or you notice your sleep schedule slipping, check in with your pharmacist or doctor first.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
2 of 2Tasimelteon also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
In vitro evidence suggests that grape seed extract might inhibit CYP3A4 enzymes. However, evidence from animal research shows that grape seed extract may induce CYP3A4 in the liver. So far, these interactions have not been reported in humans.

Your body relies on a liver enzyme called CYP3A4 to clear tasimelteon (Hetlioz) from your system. Grape seed extract might tinker with this same enzyme, but here's the honest truth: the lab studies pull in opposite directions. Some test-tube work hints it could slow CYP3A4 down (which could let tasimelteon build up, possibly leaving you groggier or more drowsy the next morning), while animal studies suggest it could speed the enzyme up (which might weaken the drug and make it less effective for resetting your sleep clock).
The key point: this has never actually been seen in real people, and it comes only from preliminary lab and animal work. Eating grapes as food is very unlikely to matter. Concentrated supplements are the wiser thing to run past your pharmacist or doctor first.

What Is Tasimelteon Used For?
Brand names: Hetlioz, Hetlioz Lq
Tasimelteon capsules are approved to treat Non-24-Hour Sleep-Wake Disorder (Non-24) in adults. Non-24 is a condition where the body's internal clock does not align with the normal 24-hour day, causing sleep and wake times to shift continuously.
Hetlioz capsules are also approved to treat nighttime sleep disturbances in Smith-Magenis Syndrome (SMS) in patients 16 years of age and older. Hetlioz LQ oral suspension is approved specifically for nighttime sleep disturbances in SMS in pediatric patients ages 3 to 15 years.
