Aspirin, Caffeine, Hydrocodone falls under CNS DEPRESSANTS — the group Hops interacts with.
Some animal research shows that hops has sedative effects.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
This combination product (Damason-P) contains hydrocodone, an opioid pain reliever that already causes drowsiness on its own. Hops are used as a mild sleep and relaxation aid, and some animal studies suggest they have a calming, sedative effect. So the concern here is simple: stacking a sedating supplement on top of an opioid could add up to more sleepiness, dizziness, slowed thinking, or feeling foggy than you'd expect from the medicine alone.
I'll be honest that this is a theoretical interaction based mostly on how hops behave, not on strong human studies, so it's not a reason to panic. Still, until you talk with your pharmacist or doctor, be cautious about driving or anything that needs you fully alert if you use both together.

CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES
2 of 3Aspirin, Caffeine, Hydrocodone also falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES.
In vitro research suggests that flavonoid constituents of hops inhibit CYP1A2 enzyme activity. However, a pharmacokinetic study in healthy postmenopausal patients shows that taking a standardized extract of spent hops containing prenylated phenols, as 59.5 mg twice daily for 2 weeks, does not affect levels of caffeine, a CYP1A2 probe substrate.

Good news here: this is a very minor, mostly theoretical concern with your Damason-P combo (aspirin, caffeine, and hydrocodone). In a test tube, certain hops flavonoids look like they might slow down the CYP1A2 enzyme, and that enzyme is what clears the caffeine piece of this product. If caffeine built up, you could in theory feel a little more jittery, restless, or have trouble winding down.
But here's the reassuring part: an actual human study using a standardized hops extract found no real change in caffeine levels. So in everyday use this pairing is unlikely to cause problems. One practical note though, both hops and the hydrocodone in this medicine can make you sleepy, so you may feel extra drowsy together. If you notice that, give your pharmacist or doctor a quick call.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

Animal research suggests that specific constituents of hops, called lupulones, can induce hepatic CYP3A4 enzyme activity. However, a pharmacokinetic study in healthy postmenopausal patients with normal metabolism shows that taking a standardized extract of spent hops containing prenylated phenols, as 59.5 mg twice daily for 2 weeks, decreases the concentration of alprazolam, a CYP3A4 probe substrate, by 7.6%. This reduction is unlikely to be clinically relevant.

If you're taking this combo pill (which packs aspirin, caffeine, and the opioid hydrocodone) and you're curious about adding hops for sleep, here's the honest picture. The concern on file is that compounds in hops might gently speed up a liver enzyme called CYP3A4, which helps process hydrocodone. In theory, that could trim its levels a bit and make the pain relief feel slightly weaker. But this is based mostly on lab and animal work, and a human study found only about an 8% change with a related medication, which is too small to really notice.
So for most folks this is a minor, mostly theoretical issue. Hops can also add to drowsiness from the hydrocodone, so go easy if you feel extra sleepy. Always run new supplements by your pharmacist or doctor first.
