nefazodone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Melatonin interacts with.

Theoretically, melatonin might increase levels of drugs metabolized by CYP3A4.
Laboratory research shows that certain lots of melatonin inhibit CYP3A4. Theoretically, combined administration of melatonin with certain drugs metabolized by CYP3A4 may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.

Here's the gist for your nefazodone: your liver clears it using an enzyme called CYP3A4. In some lab testing, certain batches of melatonin slowed that same enzyme down a little. If that happened in the body, nefazodone could clear a bit slower and build up slightly, which might mean more of its usual effects like drowsiness, dizziness, dry mouth, or lightheadedness when you stand up.

I want to be honest about how soft this is. The concern comes only from test-tube research, not real patients, and it's rated a minor, possible interaction. Both can make you sleepy on their own, so that overlap is the more practical thing to watch. Before pairing them, just run it by your pharmacist or doctor.

The evidence is strongest for certain circadian (body-clock) disorders and is more mixed for everyday insomnia. It is generally well tolerated for short-term use, but talk with your pharmacist or doctor before starting, especially for children or during pregnancy.

What Is Nefazodone Used For?

Brand name: Serzone

Nefazodone Hydrochloride tablets are approved to treat depression, specifically major depressive disorder (MDD). Clinical trials established its effectiveness over 6 to 8 weeks in outpatients and 6 weeks in hospitalized patients with depression.

Nefazodone has also been shown to help prevent depression from coming back in patients who responded well to initial treatment. Because of a serious risk of liver failure, prescribers are advised to consider other antidepressants before turning to nefazodone.
