CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
Why this applies to Rifampin
Rifampin falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Melatonin interacts with.
Laboratory research shows that certain lots of melatonin inhibit CYP3A4. Theoretically, combined administration of melatonin with certain drugs metabolized by CYP3A4 may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist: some lab studies hint that melatonin might block an enzyme called CYP3A4, which is one of the pathways your body uses to process certain medications. In theory, that could let those drugs build up a little. With rifampin, though, this is very unlikely to be a real-world problem, and honestly it's a bit ironic. Rifampin is famous for being a powerful inducer of CYP3A4, meaning it revs that enzyme up, not the other way around.
This interaction is only theoretical and comes from lab dishes, not people, so I wouldn't lose sleep over it. If anything, rifampin may speed up how fast your body clears melatonin, possibly making it less helpful for sleep. Nothing to change on your own, but feel free to run it by your pharmacist or doctor.

Melatonin is a hormone your body makes naturally to help control your sleep-wake cycle, and the supplement form is widely used to help with sleep timing problems and jet lag. The evidence is strongest for certain circadian (body-clock) disorders and is more mixed for everyday insomnia. It is generally well tolerated for short-term use, but talk with your pharmacist or doctor before starting, especially for children or during pregnancy.

What Is Rifampin Used For?
Brand names: Rifadin, Rimactane, Rifadin Injection, Rofactor
Rifampin treats all forms of tuberculosis (TB). It is almost always used in combination with other antibiotics — your doctor will build a multi-drug regimen tailored to your infection. Treatment usually runs for at least 6 months total.
Rifampin is also used to clear Neisseria meningitidis bacteria from the nose and throat in people who carry the germ without being sick themselves. It is not used to treat active meningococcal disease. The intravenous (IV) forms — Rifadin IV, Rifadin IV NOVAPLUS, and the injectable form of Rifampin — are specifically indicated when a patient cannot take the drug by mouth.
