Melatonin + Baclofen
Baclofen brand names: Lioresal, Ozobax, Lyvispah, Fleqsuvy.
Melatonin has a moderate interaction with Baclofen. Theoretically, taking melatonin with drugs that lower the seizure threshold might increase the risk of seizure activity.

Here is the interaction we found between these two, with the evidence behind it.
SEIZURE THRESHOLD LOWERING DRUGS
Why this applies to Baclofen
Baclofen falls under SEIZURE THRESHOLD LOWERING DRUGS — the group Melatonin interacts with.
Some clinical evidence suggests that melatonin may increase the frequency of seizures in certain patients, particularly children with neurological disabilities.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the short version on pairing melatonin with baclofen. This isn't about melatonin changing how your body breaks baclofen down. Instead, the concern is that both can play a role in the brain's seizure threshold. Baclofen is grouped with medicines that may slightly lower that threshold (this can show up especially if a dose is suddenly stopped), and there's some signal that melatonin may bump up seizure frequency in certain people, mainly children with neurological conditions.
Honestly, this is a theoretical concern based mostly on how these work, not strong proof, so most adults using both for sleep and muscle stiffness do fine. Still, if you or your child has a seizure history, it's worth a quick check with your pharmacist or doctor before combining them. Never stop baclofen on your own.
What These Ratings Mean
Every interaction above is graded three ways — here’s the key to each label.
Clinically significant. Avoid the combination, or use only under close professional supervision.
Usually worth avoiding or managing — may need monitoring, a dose change, or timing apart.
Generally not serious. Be aware of it and mention it to your pharmacist.

- ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
- PossibleInteraction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
- UnlikelyInteraction has been demonstrated in animal or in lab research but has been shown not to occur in humans.

About Melatonin
What is Melatonin?
- Trouble falling asleep (insomnia)
- Jet lag
- Delayed sleep phase syndrome
- Non-24-hour sleep-wake disorder
- Shift-work sleep problems
- Sleep problems in some children with conditions like autism or ADHD
Melatonin is a hormone your body makes naturally to help control your sleep-wake cycle, and the supplement form is widely used to help with sleep timing problems and jet lag. The evidence is strongest for certain circadian (body-clock) disorders and is more mixed for everyday insomnia. It is generally well tolerated for short-term use, but talk with your pharmacist or doctor before starting, especially for children or during pregnancy.
About Baclofen
What Is Baclofen Used For?
Brand names: Lioresal, Ozobax, Lyvispah, Fleqsuvy
Baclofen treats spasticity — the stiffness, muscle spasms, and rigidity that can come from neurological conditions. Oral forms (tablets, solutions, suspension, and granules) are used for spasticity from multiple sclerosis and spinal cord injuries or diseases. They are not intended for muscle spasm caused by rheumatic conditions like arthritis.
Intrathecal baclofen — delivered directly into the fluid around the spinal cord through an implanted pump — is reserved for severe spasticity that hasn't responded to oral baclofen, or when oral doses cause intolerable side effects. Gablofen (intrathecal injection) is specifically indicated for adults and children age 4 and older with spasticity of spinal or cerebral origin.
