Why this applies to Chloroquine
Chloroquine falls under SEIZURE THRESHOLD LOWERING DRUGS — the group Melatonin interacts with.
Some clinical evidence suggests that melatonin may increase the frequency of seizures in certain patients, particularly children with neurological disabilities.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the connection worth knowing if you take chloroquine along with melatonin. Chloroquine, like some other medicines, can slightly lower the brain's seizure threshold, meaning it can make seizures a bit more likely in people who are prone to them. Melatonin has also been linked, in some reports, to more frequent seizures, mostly in children with existing neurological conditions. So in theory, using both together could add up and raise that risk a little.
I want to be honest though: this is a theoretical concern based on how these work, not something proven in most people. For a typical adult with no seizure history, it's very unlikely to be a problem. Still, if you or your child has epilepsy or a seizure disorder, please check with your pharmacist or doctor first.

Melatonin is a hormone your body makes naturally to help control your sleep-wake cycle, and the supplement form is widely used to help with sleep timing problems and jet lag. The evidence is strongest for certain circadian (body-clock) disorders and is more mixed for everyday insomnia. It is generally well tolerated for short-term use, but talk with your pharmacist or doctor before starting, especially for children or during pregnancy.

What Is Chloroquine Used For?
Brand names: Aralen Phosphate, Aralen Phosphate Injection, Aralen HCL, Avloclor, Malarivon, Nivaquine
Chloroquine Phosphate tablets are used to treat uncomplicated malaria caused by susceptible strains of P. falciparum, P. malariae, P. ovale, and P. vivax. They are also used to prevent malaria when traveling to areas where chloroquine-resistant strains are not present.
Chloroquine Phosphate tablets are also approved for extraintestinal amebiasis — an infection with Entamoeba histolytica that has spread outside the intestines, such as to the liver. Important limitations apply: these tablets are not for complicated malaria, not for use in regions where chloroquine resistance is widespread, and they do not prevent malaria relapses caused by P. vivax or P. ovale without an additional medicine.
