Valacyclovir falls under SEIZURE THRESHOLD LOWERING DRUGS — the group Melatonin interacts with.
Some clinical evidence suggests that melatonin may increase the frequency of seizures in certain patients, particularly children with neurological disabilities.

Here's the honest picture with valacyclovir (Valtrex) and melatonin. This isn't about how your body breaks down the medicine. Instead, both can, in rare cases, nudge the brain toward seizure activity. Valacyclovir very uncommonly causes neurologic side effects like confusion or, rarely, seizures, usually in people with kidney problems or those taking high doses. Some weak evidence suggests melatonin may raise seizure frequency in certain people, mainly children with existing neurological conditions.
For most healthy adults taking a short course of valacyclovir for shingles or herpes, this is a theoretical concern, not something I'd expect to happen. If you have a seizure history, kidney issues, or you're giving this to a child, check with your pharmacist or doctor first.

Melatonin is a hormone your body makes naturally to help control your sleep-wake cycle, and the supplement form is widely used to help with sleep timing problems and jet lag. The evidence is strongest for certain circadian (body-clock) disorders and is more mixed for everyday insomnia. It is generally well tolerated for short-term use, but talk with your pharmacist or doctor before starting, especially for children or during pregnancy.

What Is Valacyclovir Used For?
Brand name: Valtrex
Valacyclovir tablets are used in adults to treat cold sores (herpes labialis), genital herpes (initial and recurrent episodes), and shingles (herpes zoster). They can also be taken daily as suppressive therapy to reduce how often genital herpes outbreaks occur, and to lower the risk of passing genital herpes to a partner.
In children and adolescents, valacyclovir tablets are used to treat cold sores in those aged 12 and older, and chickenpox in those aged 2 to under 18. Valacyclovir has not been established as safe and effective for most uses in immunocompromised patients, with the exception of suppressive therapy for genital herpes in HIV-infected adults.
