Why this applies to Vortioxetine
Vortioxetine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Melatonin interacts with.
Laboratory research suggests that certain lots of melatonin inhibit CYP2D6. Theoretically, combined administration of melatonin with certain drugs metabolized by the CYP2D6 enzyme may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the basic idea with these two. Your liver uses an enzyme called CYP2D6 to help clear vortioxetine (Trintellix) out of your system. Some lab studies hint that melatonin might slow that enzyme down a bit. If that happened in real life, vortioxetine could build up slightly and you might notice more of its usual effects, like nausea, headache, dizziness, or feeling more drowsy than usual.
I want to be straight with you, though: this is theoretical and only based on test-tube research, not on actual people. It's rated a minor interaction, and I wouldn't lose sleep over it. Still, since both can affect how alert you feel, it's worth a quick chat with your pharmacist or doctor before you pair them, just so they can keep an eye on how you respond.
