Diphenhydramine falls under CNS DEPRESSANTS — the group Melatonin interacts with.
Melatonin has sedative effects. Theoretically, concomitant use of melatonin with alcohol, benzodiazepines, or other sedative drugs might cause additive sedation.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the simple version: both melatonin and diphenhydramine (Benadryl) can make you drowsy on their own. Diphenhydramine is already a sedating antihistamine, which is exactly why some people use it to help them sleep. Melatonin sends your brain an "it's nighttime" signal. Stack the two together and the sleepy effects can pile up, so you might feel groggier, more dizzy, or more "out of it" than you expected, especially the next morning.
To be fair, this is a theoretical concern based on how the two work, not a dangerous reaction proven in studies. Still, I'd be careful about driving or anything needing sharp focus, and watch for next-day grogginess in older adults. If you're thinking about using both regularly, check with your pharmacist or doctor first.

SEIZURE THRESHOLD LOWERING DRUGS

Some clinical evidence suggests that melatonin may increase the frequency of seizures in certain patients, particularly children with neurological disabilities.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the deal with pairing melatonin and diphenhydramine (Benadryl): this is a theoretical concern based on how the two work, not something proven to happen in most people. Diphenhydramine is one of those drugs that can, in certain situations, slightly lower the seizure threshold (basically making seizures a touch easier to trigger). Some limited evidence suggests melatonin might raise seizure frequency in a few sensitive folks, especially children with neurological conditions.
For the average healthy adult taking occasional Benadryl for allergies or sleep, this combo isn't likely to cause a problem. But if you or your child has a seizure history or a neurological disorder, I'd run it by your pharmacist or doctor first. Also remember both can make you very drowsy, so expect extra grogginess.

Diphenhydramine temporarily relieves symptoms of hay fever, other upper respiratory allergies, and the common cold — including runny nose, sneezing, itchy or watery eyes, and itching of the nose or throat.
After Bite Xtra gel is formulated for topical use and is approved to temporarily relieve pain and itching from minor burns, minor cuts and scrapes, insect bites, minor skin irritations, and rashes from poison ivy, poison oak, and poison sumac.
