Glyburide falls under ANTIDIABETES DRUGS — the group Melatonin interacts with.

Theoretically, taking melatonin with antidiabetes drugs might increase the risk of hypoglycemia.
Some clinical research shows that melatonin reduces levels of fasting blood glucose and improves glycemic control. However, other research suggests that melatonin might impair glucose utilization and increase insulin resistance, while other research has found no effect on glucose levels. Until more is known, use melatonin cautiously in combination with antidiabetes drugs.

Here's the short version: glyburide already does the heavy lifting of lowering your blood sugar by nudging your pancreas to release more insulin. Melatonin can have its own small, somewhat unpredictable effect on blood sugar. In some studies it lowered fasting glucose, which (added on top of glyburide) could tip you toward low blood sugar, meaning shakiness, sweating, dizziness, confusion, or a racing heart. Other studies showed the opposite or no real change, so honestly the picture is mixed.

This isn't a reason to panic. The evidence is fairly weak and the effect is usually modest. If you take melatonin while on glyburide, just check your blood sugar a bit more closely at first and let your pharmacist or doctor know.

Melatonin is a hormone your body makes naturally to help control your sleep-wake cycle, and the supplement form is widely used to help with sleep timing problems and jet lag. The evidence is strongest for certain circadian (body-clock) disorders and is more mixed for everyday insomnia. It is generally well tolerated for short-term use, but talk with your pharmacist or doctor before starting, especially for children or during pregnancy.

What Is Glyburide Used For?

Brand names: Diabeta, Micronase, Glynase, Glycron, Micronized, Glynase PresTab, Albert Glyburide

Glyburide tablets are used alongside diet and exercise to help control blood sugar in adults with type 2 diabetes. It is not for type 1 diabetes or diabetic ketoacidosis (a serious complication where the body produces dangerous levels of acids called ketones).

Glyburide may be used alone or combined with metformin when metformin alone is not enough. It can also be used to help transition patients who are moving from insulin or other diabetes pills to a new regimen, under a doctor's guidance.
