Here's the simple version: both melatonin and Ingrezza (valbenazine) can make you drowsy. Valbenazine already lists sleepiness and tiredness as common side effects, and melatonin nudges your brain toward sleep too. Put them together and that drowsy feeling may just add up, leaving you groggier than expected, especially the next morning.

I want to be honest that this is a theoretical concern based on how each one works, not something proven in strong studies. For many people it's a non-issue. Laboratory research suggests that certain lots of melatonin inhibit CYP2D6. Theoretically, combined administration of melatonin with certain drugs metabolized by the CYP2D6 enzyme may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.

Here's the gist for your Ingrezza (valbenazine) and melatonin question. Part of how your body clears valbenazine relies on a liver enzyme called CYP2D6. Some lab samples of melatonin have shown they can mildly slow that enzyme down. In theory, that could let a bit more valbenazine build up in your system, which might mean stronger effects like drowsiness, fatigue, or feeling off balance.

Here's the gist if you take Ingrezza (valbenazine) and reach for melatonin at night. Your body uses an enzyme called CYP3A4 to help clear valbenazine. Some lab studies suggest certain batches of melatonin may slow that enzyme down a bit. In theory, that could let a little more valbenazine build up, which might nudge up side effects like sleepiness, dizziness, or a slow or irregular heartbeat.

What Is Valbenazine Used For?

Brand name: Ingrezza

Valbenazine — sold as Ingrezza and INGREZZA Sprinkle — is approved for two conditions in adults. The first is tardive dyskinesia, a movement disorder that causes uncontrolled, repetitive facial or body movements, often brought on by long-term use of certain psychiatric medications. The second is chorea associated with Huntington's disease, which refers to the involuntary, dance-like jerking movements that are a hallmark of that condition.

Both Ingrezza and INGREZZA Sprinkle are approved for both of these uses.
