Amlodipine falls under ANTIHYPERTENSIVE DRUGS — the group Melatonin interacts with.
Some clinical research suggests that taking melatonin decreases blood pressure in healthy adults. Also, melatonin seems to lower systolic and diastolic blood pressure in individuals with high blood pressure at nighttime or untreated essential hypertension. However, melatonin seems to worsen blood pressure in patients who are taking antihypertensive medications. Immediate-release melatonin 5 mg at night in combination with nifedipine GITS (Procardia XL) increases systolic blood pressure an average of 6.5 mmHg, diastolic blood pressure by an average of 4.9 mmHg, and heart rate by 3.9 bpm. Also, results from animal research suggest that melatonin reduces the effectiveness of certain antihypertensive drugs, including methoxamine and clonidine.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's something a little surprising about melatonin and your amlodipine. You might expect a sleep aid that gently lowers blood pressure to add to amlodipine's effect, but the research actually points the other way. In people already on blood pressure medicine, melatonin may slightly blunt how well that medicine works. In one study using a similar calcium channel blocker, nightly melatonin nudged blood pressure and heart rate up a bit rather than down.
For you, that could mean your amlodipine doesn't control your numbers quite as smoothly. This is on the milder side and only possible, not guaranteed. You don't need to stop anything, but check with your pharmacist or doctor before adding melatonin, and keep an eye on your home blood pressure readings.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
2 of 2Amlodipine also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
Laboratory research shows that certain lots of melatonin inhibit CYP3A4. Theoretically, combined administration of melatonin with certain drugs metabolized by CYP3A4 may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for these two together. Your body clears amlodipine using a liver enzyme called CYP3A4. In lab dish studies, some batches of melatonin appeared to slow that same enzyme down a bit. In theory, that could let a little more amlodipine linger in your system, which might nudge its effects up. With amlodipine, that would show up as things like ankle swelling, flushing, lightheadedness, or feeling a bit dizzy when standing.
I want to be honest about the strength of this, though. It's rated minor and comes only from test-tube and animal research, not from real patients. For most people taking both, nothing dramatic happens. Still, if you notice new dizziness or puffy ankles, mention it to your pharmacist or doctor before changing anything.

What Is Amlodipine Used For?

Amlodipine treats high blood pressure (hypertension) in adults and children aged 6–17 years. Lowering blood pressure reduces the risk of strokes and heart attacks. Sdamlo (oral solution) is specifically indicated for hypertension in adults and pediatric patients 6 years of age and older.
Amlodipine also treats coronary artery disease in adults, including chronic stable angina (chest pain with exertion), vasospastic angina (Prinzmetal's or variant angina, caused by coronary artery spasms), and documented coronary artery disease in patients without heart failure or significantly reduced heart pumping function.
