Rucaparib Camsylate falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Melatonin interacts with.

Theoretically, melatonin might increase levels of drugs metabolized by CYP2D6.
Laboratory research suggests that certain lots of melatonin inhibit CYP2D6. Theoretically, combined administration of melatonin with certain drugs metabolized by the CYP2D6 enzyme may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.

Here's the basic idea: some lab batches of melatonin can mildly block an enzyme called CYP2D6, which is one of the pathways your body uses to clear rucaparib (Rubraca). In theory, slowing that pathway could let rucaparib levels drift a little higher, which in turn could mean more of its usual side effects, things like nausea, fatigue, low blood counts, or changes in taste.

The good news is this is a very weak, theoretical concern. It comes only from test-tube research, not from real patients, and the interaction is rated minor. Rucaparib is its own active drug (not a prodrug), so the direction here would be a possible small increase in effect, not a loss of it. Still, since cancer medicines deserve care, just let your oncology team or pharmacist know before adding melatonin.

Melatonin is a hormone your body makes naturally to help control your sleep-wake cycle, and the supplement form is widely used to help with sleep timing problems and jet lag. The evidence is strongest for certain circadian (body-clock) disorders and is more mixed for everyday insomnia. It is generally well tolerated for short-term use, but talk with your pharmacist or doctor before starting, especially for children or during pregnancy.

What Is Rucaparib Used For?

Brand name: Rubraca

Rubraca (rucaparib) is used for two types of cancer in adults who carry a harmful BRCA gene mutation (either inherited or found in the tumor). First, it is used as maintenance treatment — meaning ongoing treatment to keep cancer from coming back — in people with recurrent epithelial ovarian cancer, fallopian tube cancer, or primary peritoneal cancer who responded (fully or partially) to platinum-based chemotherapy.

Second, Rubraca treats metastatic castration-resistant prostate cancer (advanced prostate cancer that has stopped responding to hormone-lowering treatment) in adults who have already received androgen receptor-directed therapy. Patients must be tested with an FDA-approved companion diagnostic to confirm the BRCA mutation before starting Rubraca for prostate cancer.
