Simeprevir Sodium falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Melatonin interacts with.

Theoretically, melatonin might increase levels of drugs metabolized by CYP3A4.
Laboratory research shows that certain lots of melatonin inhibit CYP3A4. Theoretically, combined administration of melatonin with certain drugs metabolized by CYP3A4 may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.

Here's the short version on combining melatonin with simeprevir (Olysio). Your liver uses an enzyme called CYP3A4 to help process simeprevir. Some lab samples of melatonin appear to slow that same enzyme down a bit. If that happened in the body, simeprevir could clear a little more slowly and its levels might creep up, which in theory could mean more of its usual side effects like nausea, itching, a rash, or skin that's more sensitive to sunlight.

The important thing to know is how thin this evidence is. This is only a test-tube finding, not something seen in real patients, and the interaction is rated minor. For most people taking melatonin occasionally for sleep, it's unlikely to cause a noticeable problem. Still, since simeprevir is part of a serious hepatitis C regimen, just give your pharmacist or doctor a heads-up before you start it.

What Is Simeprevir Used For?

Brand name: Olysio

  • Simeprevir is used along with ribavirin (Copegus, Rebetol) and peginterferon alfa (Pegasys) to treat chronic hepatitis C (an ongoing viral infection that damages the liver). Simeprevir is in a class of medications called protease inhibitors. It works by decreasing the amount of hepatitis C virus (HCV) in the body. Simeprevir may not prevent the spread of hepatitis C to other people.
