Oliceridine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Melatonin interacts with.

Theoretically, melatonin might increase levels of drugs metabolized by CYP2D6.
Laboratory research suggests that certain lots of melatonin inhibit CYP2D6. Theoretically, combined administration of melatonin with certain drugs metabolized by the CYP2D6 enzyme may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.

Here's the gist for Olinvyk (oliceridine), a strong IV opioid given for short-term pain in hospital settings. Some of it is cleared by an enzyme called CYP2D6. Lab studies hint that certain batches of melatonin may slow that enzyme down a bit. In theory, that could let oliceridine build up slightly, nudging up its usual effects like drowsiness, slowed breathing, low blood pressure, nausea, or constipation.

I want to be straight with you though: this is theoretical, based only on test-tube work, and it's rated a minor concern. Oliceridine is also dosed and watched closely by your care team in the hospital, so they'd catch and adjust for any extra sedation. If you take melatonin at home, just mention it to your doctor or pharmacist so it's on their radar.

Here's the situation with these two. Oliceridine (Olinvyk) is a strong IV opioid painkiller, and your liver partly clears it using an enzyme called CYP3A4. Some lab studies hint that certain batches of melatonin can slow that same enzyme down. In theory, if melatonin slows the breakdown of oliceridine, a bit more could linger in your system, which might nudge up its effects like drowsiness, slowed breathing, low blood pressure, or constipation.

I want to be honest though: this is based only on test-tube research, not real patients, and it's rated a minor interaction. On top of that, oliceridine is given in a hospital where you're closely watched. Still, since both can make you sleepy, it's worth mentioning melatonin use to your care team so they keep an eye on you.

What Is Oliceridine Injection Used For?

Brand name: Olinvyk

Olinvyk (oliceridine) injection is approved for adults to manage acute pain that is severe enough to require an intravenous opioid analgesic. It is used only when alternative treatments are ineffective, not tolerated, or otherwise unable to provide adequate pain control.

Because of serious risks including addiction, misuse, overdose, and death, Olinvyk is reserved for situations where other pain management options are not sufficient. It is not intended for routine or chronic pain management.
