Amitriptyline, Perphenazine falls under ANTICHOLINERGIC DRUGS — the group Purple Nut Sedge interacts with.

In vitro, purple nut sedge dose-dependently inhibits acetylcholinesterase (AChE). Theoretically, concurrent use of anticholinergic drugs and purple nut sedge might decrease the effectiveness of purple nut sedge or the anticholinergic agent.
Some anticholinergic drugs include atropine, benztropine (Cogentin), biperiden (Akineton), procyclidine (Kemadrin), and trihexyphenidyl (Artane).

Here's the gist with this combo. Your medicine pairs amitriptyline with perphenazine, and both of those have strong anticholinergic activity, meaning they block a brain and body messenger called acetylcholine. That's partly why they can cause dry mouth, constipation, blurry vision, and drowsiness. Purple nut sedge, at least in lab dishes, seems to do the opposite by helping that messenger stick around longer.

So in theory the two could tug against each other, possibly blunting a bit of either one's effect. Honestly though, this is only test-tube and animal data, so we really don't know if it matters in real people. I wouldn't lose sleep over it, but since your medication is a serious one, please run it by your pharmacist or doctor before adding the supplement.
