Why this applies to Estradiol

Estradiol falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Schizonepeta interacts with.

Animal research suggests that schizonepetin, a monoterpene constituent of schizonepeta, inhibits cytochrome P450 (CYP) 1A2. Theoretically, schizonepeta might increase the effects and side effects of CYP1A2 substrates.
Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), theophylline, zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the short version: your body uses an enzyme called CYP1A2 to help break down estradiol. Lab and animal studies hint that a compound in Schizonepeta may slow that enzyme down a little. If clearance slows, estradiol could linger longer and its effects (and side effects) might be slightly stronger. With estrogen, that can mean things like breast tenderness, bloating, nausea, headaches, or spotting.

That said, I want to keep this in perspective. This is only a theoretical concern based on test-tube and animal work, not human studies, so it's far from certain. Vaginal estradiol in particular puts very little into the bloodstream, which lowers the worry even more. Still, it's worth a quick chat with your pharmacist or doctor before combining them, just so someone who knows your full picture can weigh in.

Animal research suggests that schizonepetin, a monoterpene constituent of schizonepeta, induces cytochrome P450 (CYP) 3A4. Theoretically, schizonepeta might decrease the effects of CYP3A4 substrates.
Some substrates of CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.

Here's the gist for estradiol: your body clears this hormone partly through a liver enzyme called CYP3A4. Animal studies hint that a compound in Schizonepeta may speed that enzyme up. If that holds true in people, your system could break down estradiol a little faster, which might leave you with weaker results, things like returning hot flashes, vaginal dryness, or itching that the estrogen was helping to settle.

That said, this is only a theoretical concern based on lab and animal work, not human studies, so I wouldn't lose sleep over it. Low-dose vaginal estradiol especially works mostly right where it's applied. Still, it's worth a quick chat with your pharmacist or doctor before adding this herb.
