Why this applies to Venlafaxine
Venlafaxine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Shiitake Mushroom interacts with.
In vitro studies suggest that the shiitake mushroom extract AHCC might induce the CYP2D6 enzyme. This effect has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for Effexor (venlafaxine). Your body relies on a liver enzyme called CYP2D6 to help process this medication. Some lab studies hint that concentrated shiitake extracts (like AHCC) might speed that enzyme up a bit. If that happened, your body could clear venlafaxine a little faster, which in theory could make it work less well, so you might notice your depression or anxiety symptoms creeping back.
That said, I want to keep this in perspective. This is only a test-tube finding, and it has never actually been shown in people. Eating shiitake as food is not a concern here. If you're thinking about a strong shiitake supplement, just run it by your pharmacist or doctor first so they can keep an eye on how you're feeling.

Extended-release tablets are approved for major depressive disorder and social anxiety disorder. Immediate-release venlafaxine tablets are indicated for major depressive disorder only.
