Lapatinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Sulforaphane interacts with.

Theoretically, sulforaphane might increase the levels and effects of CYP3A4 substrates.
Animal and in vitro research shows that sulforaphane inhibits CYP3A4 enzyme activity and downregulates its expression. This interaction has not been reported in humans.

Here's the gist for Tykerb (lapatinib): your liver relies heavily on an enzyme called CYP3A4 to clear this drug from your body. In lab and animal studies, sulforaphane appears to slow that enzyme down. If that same effect happened in people, lapatinib could build up to higher levels than intended, which might mean more of its usual side effects, things like diarrhea, nausea, rash, fatigue, or in rarer cases liver or heart strain.

I want to be honest about the strength of this: it's only theoretical right now and has not been seen in actual patients. The broccoli on your dinner plate is very unlikely to matter. A concentrated sulforaphane supplement is a different story, so please run it by your oncologist or pharmacist before adding one.

What Is Lapatinib Used For?

Brand name: Tykerb

Lapatinib tablets — sold under the brand name Tykerb — are used to treat advanced or metastatic breast cancer in patients whose tumors overexpress the HER2 protein. They are always used in combination with another medicine, not alone.

Tykerb is approved in combination with capecitabine for HER2-positive advanced or metastatic breast cancer in patients who have already received prior treatment including an anthracycline, a taxane, and trastuzumab, and whose disease has progressed on trastuzumab. Tykerb is also approved in combination with letrozole for postmenopausal women with hormone receptor-positive, HER2-positive metastatic breast cancer when hormonal therapy is appropriate.
