CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES

Oestrogen falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Sulforaphane interacts with.
Animal and in vitro research shows that sulforaphane inhibits CYP1A2 enzyme activity. However, its precursor glucoraphanin also appears to increase the expression of the CYP1A2 enzyme, which could lead to increased metabolism of CYP1A2 substrates. These effects have not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the short version: your body uses an enzyme called CYP1A2 to help break down estrogen. Lab and animal studies suggest sulforaphane (the compound concentrated from broccoli) can block that enzyme, which in theory could let estrogen build up a bit and make its effects, and side effects, like breast tenderness, bloating, nausea, or headaches, more noticeable. To muddy things further, a related broccoli compound (glucoraphanin) may actually speed up that same enzyme, which would push estrogen levels the other way.
So honestly, the direction isn't clear, and none of this has been confirmed in real people. Eating broccoli as food almost certainly won't matter. If you're considering a concentrated sulforaphane supplement, just run it by your pharmacist or doctor first.

Animal and in vitro research shows that sulforaphane inhibits CYP3A4 enzyme activity and downregulates its expression. This interaction has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Your body uses an enzyme called CYP3A4 to help break down estrogen and clear it out. In lab and animal studies, sulforaphane (the broccoli compound) appears to slow that enzyme down. If that held true in people, estrogen could clear a little more slowly and build up, which might mean stronger effects or more of the usual estrogen side effects like breast tenderness, bloating, nausea, headaches, or spotting.
Here's the reassuring part: this has only been seen in test tubes and animals, never confirmed in humans, so it's theoretical. Eating broccoli and other veggies is not a concern. A concentrated sulforaphane supplement is where I'd pause. Just run it by your pharmacist or doctor before adding one.
