Amitriptyline, Perphenazine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Sweet Annie interacts with.

Sweet Annie may alter plasma levels and clinical effects of drugs metabolized by CYP3A4.
In vitro research shows that the Sweet Annie constituent artemisinin induces CYP3A4, possibly increasing CYP3A4 activity by 1.9-fold. However, Sweet Annie extract seems to inhibit the activity of CYP3A4 in vitro, suggesting that other constituents of Sweet Annie play a role in its effects on the overall activity of this enzyme. More information is needed to determine whether taking Sweet Annie extract affects the metabolism of CYP3A4 substrates.

This combo pill pairs amitriptyline (a tricyclic antidepressant) with perphenazine (an antipsychotic). Both rely partly on liver enzymes, including CYP3A4, to be cleared from your body. Sweet Annie may tug on that enzyme, but the lab data points in two directions at once: one constituent (artemisinin) seems to speed CYP3A4 up, while the whole plant extract seems to slow it down. Honestly, we don't yet know which wins out in a real person.

If clearance sped up, the medicine could work less well; if it slowed down, you might notice more drowsiness, dry mouth, constipation, dizziness, or a faster heartbeat. Right now this is only test-tube evidence, so don't be alarmed. Still, please run it by your pharmacist or doctor before combining them.
