Why this applies to Crizotinib

Crizotinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Sweet Annie interacts with.

Sweet Annie may alter plasma levels and clinical effects of drugs metabolized by CYP3A4.
In vitro research shows that the Sweet Annie constituent artemisinin induces CYP3A4, possibly increasing CYP3A4 activity by 1.9-fold. However, Sweet Annie extract seems to inhibit the activity of CYP3A4 in vitro, suggesting that other constituents of Sweet Annie play a role in its effects on the overall activity of this enzyme. More information is needed to determine whether taking Sweet Annie extract affects the metabolism of CYP3A4 substrates.

Here's the situation with Sweet Annie and your crizotinib (Xalkori). Crizotinib is cleared from your body by a liver enzyme called CYP3A4. Sweet Annie can tug on that same enzyme, but it pulls in two directions: one of its compounds, artemisinin, may speed the enzyme up (which could lower crizotinib levels and weaken its cancer-fighting effect), while the whole-plant extract seems to slow it down (which could raise levels and worsen side effects like nausea, vision changes, swelling, fatigue, or heart rhythm changes).

Honestly, this is based only on lab studies, so we don't truly know what happens in real people. But with a cancer drug, I wouldn't gamble. Please run any Sweet Annie use past your oncologist or pharmacist first.

What Is Crizotinib Used For?

Brand name: Xalkori

Xalkori (crizotinib) is used to treat adult patients with metastatic non-small cell lung cancer (NSCLC) whose tumors test positive for the ALK or ROS1 gene abnormality, as confirmed by an FDA-approved test.

Xalkori is also used to treat pediatric patients aged 1 year and older and young adults with relapsed or refractory systemic ALK-positive anaplastic large cell lymphoma (ALCL). Additionally, it treats adult and pediatric patients aged 1 year and older with unresectable, recurrent, or refractory ALK-positive inflammatory myofibroblastic tumor (IMT). Note: Xalkori's safety and effectiveness in older adults with relapsed or refractory ALK-positive ALCL have not been established.
