Entrectinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Thunder God Vine interacts with.

Theoretically, thunder god vine might increase the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that triptolide, a constituent of thunder god vine, inhibits CYP3A4 enzymes in human microsomes. This interaction has not been reported in humans.

Here's the gist for Rozlytrek (entrectinib). Your body relies on an enzyme called CYP3A4 to break this cancer drug down and keep its levels in check. Thunder God Vine contains a compound (triptolide) that, in lab dish studies, slowed that same enzyme. If it does the same thing in people, entrectinib could clear out more slowly and build up higher than intended.

More entrectinib in your system can mean stronger side effects, things like dizziness, fatigue, constipation, swelling, changes in heart rhythm, or trouble with balance. I want to be honest though: this is only a theoretical concern from test-tube work, not something actually seen in patients. Still, with a cancer medicine, don't guess. Please loop in your oncologist or pharmacist before combining the two.

What Is Entrectinib Used For?

Brand name: Rozlytrek

Rozlytrek (entrectinib) is used to treat adult patients with ROS1-positive metastatic non-small cell lung cancer (NSCLC), as confirmed by an FDA-approved test. It is also approved for adult and pediatric patients older than 1 month of age who have solid tumors — any cancer type — that carry an NTRK gene fusion detected by an FDA-approved test, provided the tumor has no known acquired resistance mutation, is metastatic or cannot be surgically removed without severe harm, and has progressed after prior treatment or has no satisfactory alternative therapy.

The NTRK gene fusion approval in solid tumors was granted under accelerated approval, meaning it is based on tumor response rates. Continued approval may depend on results from confirmatory clinical trials.
