Adagrasib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Tree Turmeric interacts with.

Theoretically, tree turmeric might increase the levels and clinical effects of drugs that are substrates of CYP3A4.
In vitro research and preliminary clinical evidence suggest that berberine, a constituent of tree turmeric, moderately inhibits CYP3A4.

Here's the gist for Adagrasib (Krazati), which is the active drug itself, not a prodrug. Your body relies on an enzyme called CYP3A4 to break it down and clear it. The berberine in tree turmeric appears to slow that enzyme down, so adagrasib could build up to higher levels in your blood than expected.

In practical terms, that may mean stronger effects and more side effects, things like nausea, diarrhea, vomiting, tiredness, or changes in your heart's QT interval (an electrical rhythm doctors watch closely with this medicine). The signal here is partly theoretical, but berberine is a known CYP3A4 blocker, so I wouldn't ignore it. Please loop in your oncology team or pharmacist before adding tree turmeric.

About Adagrasib

What Is Adagrasib Used For?

Brand name: Krazati

Krazati (adagrasib) is used to treat adults with locally advanced or metastatic non-small cell lung cancer (NSCLC) that carries a specific genetic change called a KRAS G12C mutation. It is used as a single agent for NSCLC after at least one prior treatment, and patients must be confirmed to have this mutation by an FDA-approved test.

Krazati is also used in combination with another drug called cetuximab to treat adults with KRAS G12C-mutated locally advanced or metastatic colorectal cancer (CRC) who have already been treated with specific prior chemotherapy regimens. Both uses were approved under the FDA's accelerated approval program based on tumor response data, and continued approval may depend on results from confirmatory trials.
