L-glutamine is generally well tolerated by healthy adults when taken by mouth. The most common side effects are gastrointestinal: belching, bloating, constipation, diarrhea, flatulence, nausea, vomiting, and stomach pain.

Headache and musculoskeletal pain have also been reported. Two people with bipolar disorder experienced mania or hypomania at doses up to 4 grams daily, and one person reported dizziness at a high dose (which resolved when the dose was lowered).

If you have kidney or liver disease, use this supplement only under a doctor's supervision. The safety data during pregnancy is adequate—the ingredient is rated Likely Safe.
