Sleep Tonight Ingredients & Drug Interactions
by Nature's Way
What is this page for?
First and foremost: checking Sleep Tonight against your medications. The heart of this page is the interaction checker and the full interaction report — how this product’s ingredients may interact with prescription and over-the-counter medicines you may be taking.
Around that, we add a pharmacist’s high-level view of the product as a whole — what’s inside, the evidence for its stated use, how transparent the label is, and what safety data exists — so you can see the full picture in one place. It’s educational information from our licensed clinical databases and the clinical staff at HelloPharmacist — not medical advice — and we don’t sell or endorse products. Our editorial policy

HelloPharmacist Scorecard of Sleep Tonight by Nature's Way
Our pharmacy team’s full take, with four database checks built into the cards below — a summary of what is known, not a grade of the product itself.
What’s inside
Partial disclosure
Sleep Tonight contains 6 ingredients. The active ones are L-theanine, a compound that may support relaxation; phosphatidylserine, which plays a role in brain cell health; ashwagandha (supplied as Sensoril), an herb used for stress and sleep; and three botanical extracts: honokiol from magnolia bark, epimedium aerial parts, and magnolol.
The product also includes a stress-reducing proprietary blend and a cortisol-reducing proprietary blend, whose exact components aren't detailed on the label. The tablet is made up of standard inactive ingredients like cellulose, magnesium stearate, and silica — common binding and flow agents.
Does it work?
Moderate evidence
The evidence for Sleep Tonight's ingredients is mixed. Ashwagandha has possibly effective ratings for insomnia, anxiety, stress, and generalized anxiety disorder — the main uses you'd expect from a sleep supplement.
Phosphatidylserine is possibly effective for age-related cognitive decline and Alzheimer disease, though that's not a sleep benefit. L-theanine is possibly effective for cognitive function, but evidence for anxiety, sleep, and other conditions in its monograph is insufficient.
For honokiol, epimedium, and magnolol, we hold no effectiveness data.
How safe is it?
Well-documented data
L-theanine and phosphatidylserine are generally well tolerated in the short term, though long-term safety isn't fully studied. Ashwagandha is generally well tolerated in the short term, but the data are limited.
Side effects can include headaches, drowsiness, increased sleep duration, and dream activity (from L-theanine); flatulence, nausea, gastrointestinal upset, and insomnia, especially at higher doses (from phosphatidylserine); and diarrhea, gastrointestinal upset, nausea, and vomiting (from ashwagandha, though these are uncommon at typical doses). Rare serious concerns with ashwagandha include acute liver damage and hepatitis.
Pregnancy and breastfeeding: L-theanine, phosphatidylserine, and ashwagandha all lack sufficient safety data in pregnancy and lactation — the facts advise against use during both.
Meds to double-check
Moderate interaction found
Before taking Sleep Tonight, check whether you're on blood pressure-lowering drugs (antihypertensives) — both L-theanine and ashwagandha may intensify their effects. If you take sedatives, anti-anxiety drugs, benzodiazepines, or other CNS depressants, ashwagandha and L-theanine may increase drowsiness.
Double-check if you're on diabetes medications, thyroid hormone, immunosuppressants, anticholinergic drugs, or cholinergic drugs — ashwagandha and phosphatidylserine interact with these. Run your medications through the tool below.
The bottom line
Scorecard at a glancePartially disclosed formula with some supporting evidence for its stated purpose. Moderate medication interactions have been identified, and safety information is well characterized.
Sleep Tonight may help you with sleep and stress if you don't take medications that interact with its ingredients. If you're on blood pressure drugs, sedatives, benzodiazepines, diabetes medications, thyroid replacement, or immune suppressants, talk to your pharmacist before starting — ashwagandha's interactions are the main concern.
Pregnant or breastfeeding women should skip this product. Check your exact medications on this page first.

Sleep Tonight by Nature's Way Drug Interactions
HelloPharmacist Interaction Report
Sleep Tonight by Nature's Way contains ingredients that interact with a range of medications.
The most serious concern is ashwagandha (Sensoril), which carries moderate-severity interactions with several drug classes. Ashwagandha may potentiate CNS depressants (sedatives, anxiolytics, barbiturates) and benzodiazepines, increasing drowsiness and sedation risk.
It can also lower blood pressure, adding to the effects of antihypertensive drugs, and may raise the risk of low blood sugar (hypoglycemia) if you're taking antidiabetes medications.
Read the full breakdown — every affected drug type, severity by severity
Ashwagandha also carries moderate interactions with thyroid hormone replacement, potentially increasing its effects and side effects, and with immunosuppressants, where it may reduce their effectiveness. There's a moderate risk of liver damage when ashwagandha is combined with hepatotoxic drugs.
Additionally, ashwagandha may reduce levels of certain medications metabolized by the CYP1A2 enzyme (a minor interaction).
L-theanine and phosphatidylserine add their own moderate interactions: L-theanine may lower blood pressure alongside antihypertensives, and phosphatidylserine may interfere with anticholinergic drugs or have additive effects with cholinergic medications. L-theanine also has minor interactions with CNS depressants and serotonergic drugs.
We could not check honokiol, epimedium aerial parts extract, or magnolol — interaction data isn't on file for those ingredients.
Altogether, these interactions span 1,446 individual medications. Check your exact medications with the tool on this page before starting.

Ashwagandha might increase the effects and adverse effects of thyroid hormone.
Concomitant use of ashwagandha with thyroid hormones may cause additive therapeutic and adverse effects. Preliminary clinical research and animal studies suggest that ashwagandha boosts thyroid hormone synthesis and secretion. In one clinical study, ashwagandha increased triiodothyronine (T3) and thyroxine (T4) levels by 41.5% and 19.6%, respectively, and reduced serum TSH levels by 17.4% from baseline in adults with subclinical hypothyroidism.
Cytochrome P450 1A2 (Cyp1A2) Substrates
Theoretically, ashwagandha might decrease the levels and clinical effects of CYP1A2 substrates.
In vitro research shows that ashwagandha extract induces CYP1A2 enzymes.
Cytochrome P450 3A4 (Cyp3A4) Substrates
Theoretically, ashwagandha might decrease the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that ashwagandha extract induces CYP3A4 enzymes.
Serotonergic Drugs
Some animal studies have reported that ashwagandha can enhance serotonergic transmission by altering certain serotonin (5-HT) receptors. However, there is no evidence to suggest that ashwagandha increases the risk of serotonin-related effects, and there have been no published case reports of serotonin syndrome when combined with other serotonergic drugs. Nevertheless, due to the lack of extensive studies on the matter and the fact that ashwagandha appears to affect serotonergic pathways, it would be prudent to exercise caution when combining it with drugs that affect serotonin. Epimedium Aerial Parts Extract
Anticoagulant/Antiplatelet Drugs
Theoretically, horny goat weed might increase the risk of bleeding.
In vitro research and animal research shows that horny goat weed can inhibit platelet aggregation and thrombus formation. This effect has not been reported in humans.
Antihypertensive Drugs
Theoretically, horny goat weed might increase the risk of hypotension.
Laboratory research suggests that horny goat weed might have hypotensive effects. This effect has not been reported in humans.
Cytochrome P450 1A2 (Cyp1A2) Substrates
Theoretically, horny goat weed might increase the effects and side effects of CYP1A2 substrates.
In vitro, horny goat weed leaf extract inhibits CYP1A2. This effect has not been reported in humans.
Cytochrome P450 2B6 (Cyp2B6) Substrates
Theoretically, horny goat weed might increase the effects and side effects of CYP2B6 substrates.
In vitro, horny goat weed leaf extract inhibits CYP2B6. This effect has not been reported in humans.
Cytochrome P450 3A4 (Cyp3A4) Substrates
Theoretically, horny goat weed might increase the effects and side effects of CYP3A4 substrates.
In vitro, horny goat weed extract inhibits CYP3A4 and suppresses CYP3A4 mRNA expression. This effect has not been reported in humans.
Estrogens
Theoretically, concomitant use of horny goat weed with estrogens might increase their therapeutic and adverse effects.
In vitro evidence suggests that horny goat weed has estrogenic activity. In clinical research, horny goat weed has been shown to increase blood levels of estrogen in some females.
L-Theanine
Antihypertensive Drugs
Theanine might lower blood pressure, potentiating the effects of antihypertensive drugs.
Animal research shows that theanine can lower blood pressure in spontaneously hypertensive animals. Theoretically, concomitant use of theanine and antihypertensive drugs might potentiate the antihypertensive activity.
Cns Depressants
Theoretically, theanine might have additive sedative effects when used in conjunction with CNS depressants. However, it is unclear if this concern is clinically relevant.
Theoretically, theanine may compete with glutamate and/or increase plasma gamma-aminobutyric acid (GABA) levels, which could cause CNS depression. In one clinical study, some subjects taking oral theanine reported drowsiness.
Serotonergic Drugs
Clinical studies regarding the effects of L-theanine on serotonin levels are conflicting. Some studies suggest it can increase serotonin levels in the brain while others report that it may decrease them. Nevertheless, there have been no reports of l-theanine being a causative agent in serotonergic-related side effects or serotonin syndrome.
Magnolia Bark Extract
Anticoagulant/Antiplatelet Drugs
Theoretically, magnolia might have additive effects and increase the risk of bleeding when used with anticoagulant or antiplatelet drugs.
In vitro research shows that the chemicals magnolol and honokiol, isolated from magnolia bark, inhibit platelet aggregation that is experimentally induced by collagen and arachidonic acid. However, they do not inhibit platelet aggregation that is induced by adenosine diphosphate, platelet-activating factor, or thrombin. This interaction has not been reported in humans.
Cns Depressants
Theoretically, concomitant use of large doses of magnolia bark and CNS depressants might have additive effects.
In vitro and animal research shows that constituents extracted from magnolia bark, especially honokiol and magnolol, have sedative effects. These effects may be due to the inhibition of catecholamine release and modulation of gamma-aminobutyric acid-A (GABA-A) receptors.
Phosphatidylserine
Anticholinergic Drugs
Theoretically, phosphatidylserine might decrease the effectiveness anticholinergic drugs.
Phosphatidylserine is thought to increase acetylcholine levels, which could theoretically interfere with the activity of anticholinergic agents.
Cholinergic Drugs
Theoretically, phosphatidylserine might have additive effects with cholinergic drugs.
Phosphatidylserine is thought to increase acetylcholine levels, which could theoretically lead to additive cholinergic effects when used with cholinergic drugs.
