Homatropine Methylbromide, Hydrocodone falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's the wrinkle with this combo. Hydrocodone is a prodrug in part. Your body uses an enzyme called CYP2D6 to turn some of it into hydromorphone, a stronger pain-relieving form. The beta-carboline compounds in ayahuasca (harmine and harmaline) may block CYP2D6. If that happens, less hydrocodone gets converted, so you might actually get less pain or cough relief than expected.

I'll be honest, this is theoretical. It comes only from lab studies, not real people, so I can't tell you how strong it would be in practice. Beyond this, ayahuasca is a powerful MAOI brew with its own serious risks. Please talk with your pharmacist or doctor before mixing these.

Here's the idea behind this pairing. The hydrocodone in Hycodan and Tussigon is partly processed by a liver enzyme called CYP3A4. Lab studies hint that the beta-carboline compounds in ayahuasca can block that enzyme. If that happened in real life, hydrocodone could linger longer and build up, which might mean stronger opioid effects like heavy drowsiness, slowed breathing, constipation, and dizziness. The homatropine part can add to dry mouth and blurry vision.

I want to be honest: this is only a theoretical, test-tube concern and hasn't been shown in people. Still, ayahuasca's MAOI activity can be risky with many medicines on its own, so please check with your doctor or pharmacist before combining them.
